Formaldehyde is an important chemical used widely by industry to manufacture building materials and numerous household products. Exposure to formaldehyde can result in various negative health effects. In this project, we are going to find a more cost-effective way to battle this harmful reagent. Our project is based on the work by TMU-TOKYO2012 with some adjustments. Removal of formaldehyde can occur by different in vivo pathways. Among one of them, glutatione independent formaldehyde dehydrogenase was used to oxidize and convert formaldehyde to formic acid which was further oxidized to carbon dioxide by formic acid dehydrogenase. As a metabolite is oxidized, NAD+ (nicotinamide adenine dinucleotide) accepts two electrons and a hydrogen ion (H+); this results in NADH + H+. The main intermediate product of the reaction was formic acid, and the further oxidation of it was the rate-limiting step for the degradation of fromaldehyde. We would like to construct expression vectors using formaldehyde-derived promoter for inducible expression of formaldehyde dehydrogenase, formic acid dehydrogenase and NADH dehydrogenase as well, in case of formic acid accumulation and deletion of NAD+.
